Technique

1. Chemically encode the sequence in the set of fragment lengths present in 4 primer extension reactions ( Sanger's Method)
2. Electrophorese the products and measure the bandsto obtain a 4-component time series
3. Recover the sequence by analyzing peaks in the components of this time series to determine the order of fragments present in each reaction ( basecalling)
